15-P030 Embryo patterning in gastrulating pig and mouse embryos

چکیده
Vertebrate ancestors would have increased their egg size to store yolk, and the increase is considered to have altered the cleavage pattern and germ layer formation. Amphibian holoblastic cleavage in which all blastomeres contribute to any one of the three primary germ layers has been widely thought to be a developmental pattern in the stem lineage of vertebrates, and meroblastic cleavage to have evolved independently in each vertebrate lineage. In extant primitive vertebrates, agnathan lamprey and basal bony fishes also undergo holoblastic cleavage, and their vegetal blastomeres have been generally thought to contribute to embryonic endoderm. However, the identification of their primary germ layers based on molecular evidences was not reported. We performed the marker analyses in most basal ray-finned fish bichir and agnathan lamprey embryos, resulting that their mesoderm and endoderm develop in the equatorial marginal zone, and their vegetal cell mass is extraembryonic nutritive yolk cells, having non-cell autonomous meso-endoderm inducing activity. Furthermore, Eomesodermin, but not VegT, orthologs are expressed maternally in these animals as well as zebrafish, mouse and protochordates, suggesting that VegT is a maternal factor for endoderm differentiation only in amphibian. The study raises the viewpoint that the lamprey/bichir type holoblastic development would have been ancestral to extant vertebrates and retained in their stem lineage as a preliminary state toward the meroblastic development; amphibian-type holoblastic development would have been acquired secondarily, accompanied by the exploitation of new molecular machinery such as maternal VegT.

Potential of Mouse and Human Embryonic Fibroblasts to Support Mouse Embryo Development Following Exposure to Laboratory Temperature
Purpose: Present study was designed to evaluate the potential of co-culture systems to overcome deleterious effect of exposure of mouse 2-cell embryos to low temperature. Materials and Methods: 2-cell embryos were flushed from oviduct of super ovulated NMRI mice into HTF medium with 15% BSA. After washing 3 times with HBSS and with 15% BSA, embryos were exposed to laboratory temperature (LT) (...
متن کاملLocalization and loss-of-function implicates ciliary proteins in early, cytoplasmic roles in left-right asymmetry.
Left-right asymmetry is a crucial feature of the vertebrate body plan. While much molecular detail of this patterning pathway has been uncovered, the embryonic mechanisms of the initiation of asymmetry, and their evolutionary conservation among species, are still not understood. A popular recent model based on data from mouse embryos suggests extracellular movement of determinants by ciliary mo...
متن کاملP-30: Developmental Capacity and Blastocyst Formation of Thawed Tetrahedral Versus Non-Tetrahedral 4-cell Stage Mouse Embryos After Vitrification
Background: It was reported in a literature that approximately one third of the 4-cell stage embryos did not exhibit a tetrahedral shape. Non-tetrahedral embryos showed a lower in vitro developmental potential than tetrahedral embryos. Recently vitrification technology has been widely employed for embryo cryopreservation. The objective of this study was to prove our hypothesis that vitrified - ...
متن کاملThe developmental fate of androgenetic, parthenogenetic, and gynogenetic cells in chimeric gastrulating mouse embryos.
Both a maternal and a paternal genomic contribution are necessary for completion of embryonic development in the mouse. Parthenogenetic embryos, with only a maternally inherited genome, and androgenetic embryos, with only a paternally inherited genome, fail to develop to term, and these two types of isoparental embryos fail in development in characteristic ways. In this paper we describe the co...
متن کاملP-98: Effect of Mouse Embryo Vitrification on Histone Modifications
Background: Vitrification has been usually used as an assisted reproductive technology in animals and humans. This method needs high concentrations of cryoprotectants that can be toxic with high cooling degrees. Then, vitrification could be change histone modifications such as methylation and acetylation can performance as regulatory controls of gene transcription. So, the purpose of the presen...
